ABSTRAK

Tanah, air laut, lumpur, kompos, sampah rumah tangga, dan makanan busuk
merupakan sumber mikroorganisme penghasil antibiotik. Namun sebagian besar
mikroorganisme penghasil antibiotik berasal dari mikroorganisme tanah, terutama
Actinomycetes. Actinomycetes merupakan mikroorganisme tanah yang berpotensi
menghasilkan senyawa antibakteri. Penelitian ini bertujuan untuk mendapatkan
isolat Actinomycetes penghasil antibakteri/antibiotik dari tanah di Kawasan
Temiang Sekupang. Penelitian eksperimental ini dilakukan dengan mengambil
sampel tanah dari tiga lokasi berbeda (kebun, perairan, dan hutan) di Kawasan
Temiang. Isolasi Actinomycetes dilakukan menggunakan media Nutrient Agar (NA)
dengan penambahan antibiotik Amoxicillin. Uji aktivitas antibakteri dilakukan
dengan metode difusi kertas cakram terhadap bakteri Staphylococcus aureus dan
Escherichia coli. Hasil penelitian menunjukkan diperoleh 18 isolat yang terdiri dari
9 isolat untuk masing-masing bakteri uji. Isolat A (A1, A2, A3), B (B1, B2, B3) dan
C (C1, C2, C3) menunjukkan aktivitas antibakteri kategori sedang dengan diameter
zona hambat dalam rentang 10,70 - 10,90 mm terhadap kedua bakteri uji. Hasil
pewarnaan Gram menunjukkan semua isolat merupakan bakteri Gram positif
berbentuk batang dan bulat yang merupakan karakteristik Actinomycetes. Penelitian
ini membuktikan bahwa tanah di Kawasan Temiang mengandung Actinomycetes
yang berpotensi sebagai penghasil antibakteri.
